چکیده

In the complex microenvironment where they evolve, developing cells undergo rapid programmed cell death (PCD) when cytokines that support them become limiting. The transcriptional mechanisms of cytokine-withdrawal apoptosis are poorly understood. In this report, we used early B-lymphocyte tissue culture and transgenic cells to demonstrate that nuclear factor-kB (NF-kB) promotes apoptosis during cytokine withdrawal-induced PCD. In the progenitor B lymphocyte model FL5.12, whereas NF-kB has an antiapoptotic function in response to tumor necrosis factor-a, cytokine withdrawal causes nuclear translocation of NF-kB/cRel, where it is apoptogenic. Inhibition of NF-kB activation delays cytokine withdrawal-induced PCD in both FL5.12 and transgenic early B cells. Additionally, reconstituting a bone marrow microenvironment ex vivo abrogates the differential apoptotic pattern between control and transgenic early B cells.

Early NF-kB activation is associated with hormone-induced pancreatitis

Gukovsky, Ilya, Anna S. Gukovskaya, Thane A. Blinman, Vjekoslav Zaninovic, and Stephen J. Pandol. Early NF-kB activation is associated with hormone-induced pancreatitis. Am. J. Physiol. 275 (Gastrointest. Liver Physiol. 38): G1402–G1414, 1998.—Inflammation and cell death are critical to pathogenesis of acute pancreatitis. Here we show that transcription factor nuclear factor-kB (NF-kB), which r...

Impact of Duration and Severity of Persistent Pain on Programmed Cell Death

Programmed cell death is a highly regulated form of cell death, mostly distinguished by the activation of a family of cystein-aspartate proteases (caspases) that cleave various proteins resulting in morphological and biochemical changes characteristic of this form of cell death. Several recent studies have addressed the role of programmed cell death in inflammatory and chronic pain states. Casp...

Activation of the Transcriptional Function of the NF-kB Protein c-Rel by O-GlcNAc Glycosylation

D ow nladed fr The transcription factor nuclear factor kB (NF-kB) rapidly reprograms gene expression in response to various stimuli, and its activity is regulated by several posttranslational modifications, including phosphorylation, methylation, and acetylation. The addition of O-linked b-N-acetylglucosamine (a process known as O-GlcNAcylation) is an abundant posttranslational modification tha...

Differential expression of Rel/NF-kB and octamer factors is a hallmark of the generation and maturation of dendritic cells

A key feature of maturation of dendritic cells is the down-regulation of antigenprocessing and up-regulation of immunostimulatory capacities. To study the differential expression of transcription factors in this process, we investigated the nuclear translocation and DNA binding of Rel/NF-kB and octamer factors during in vitro generation and maturation of dendritic cells compared with macrophage...
